No B button challenge. - Some tips for this are, holding items, spin jump turns into jump. If you know the level has spin blocks to get past, don't get a powerup, if you do, make sure there's a way to lose the power up.

No Power Up challenge. - Title basically explains it, don't use Power Ups.
